The in vivo and in vitro Effect of Dipyridamole ( Persantin(R) ) and Aspirin on the Platelet Aggregation

Abstract
A study was performed to evaluate the in vivo and in-vitro effect of Dipyridamole (Persantin¢ç) and Aspirin on the platelet aggregation in 4 arterial thrombosis patients, and 17 healthy volunteers, . using Chrono-Log Aggregometer. Following results were obtained.
1. After oral administration of Dipyridamole (150^¢¥300 mg/day) for 7 days, there were no changes in the ADP or epinephrine induced platelet aggregation in 4 patients with arterial thrombosis.
2. After intravenous administration of Dipyridamole (0.5- 5-0mg/kg) in healthy volunteers, there were no significant change in the ADP or, epinephrine induced platelet . aggregation and also in spontaneous aggregation.
3. To observe. the in vitro effect of Dipridamole on platelet aggregation, platelet rich plasma(PRP) obtained from 4 healthy volunteers were, tested. ADP induced platelet aggregation, was inhibited in the concentration of 25 pg/ml of- Dipyridamole in two persons, and in 50 ,ug/ml in the other two. Epinephrine induced platelet aggregation was inhibited in two person above the concentration of 75 pg/ml, but other two person¢¥s platelets were not inhibited even in 100 pg/ml.
4. 1.2 mg/kg of oral Aspirin was the minimal effective dose, which could the platelet aggregation studied in 10 healthy persons.
5. Secondary . aggregation of platelet could be inhibited in the concentration of as low as 0.8^¡¯81pg/ml of Aspirin in in-vitro study.
